生物细胞的发育分化正受到越来越多的人们的重视,细胞的有丝分裂方向是影响细胞发育分化的因素之一.有丝分裂的取向问题关系到胞内物质能量的分配问题,研究分析影响细胞分裂方向的因素则显得至关重要.本文研究了单轴拉伸应力对细胞应力纤维排布方向和细胞有丝分裂方向的影响.课题探讨当细胞黏附的基底膜拉伸引起细胞相应变形时导致的抗应力微丝沿应变方向的重新排布与其后的细胞分裂方向是否有关系.实验以MC3T3成骨细胞为主要研究对象,使用由等双轴应力拉伸装置改制的单轴应力拉伸装置进行应力加载下的细胞培养,并主要运用鬼笔环肽和DAPI荧光标记物分别对细胞微丝和细胞核进行荧光标记,观察细胞在单轴应力拉伸状况下应力纤维的排布情况和细胞有丝分裂方向.本文对细胞在单轴应力拉伸下细胞形态的变化进行了观察比较,并且分别对细胞应力纤维排布方向与单轴应力方向的关系以及细胞有丝分裂方向与单轴应力方向的关系进行了统计分析.实验结果显示细胞在8%单轴应力拉伸下形态有比较明显的变化,细胞的长轴基本延单轴应力方向排列.统计结果表明细胞应力纤维的排布方向和细胞有丝分裂方向基本与单轴应力方向保持一致.
